Eighteen years after Neukom has completed his law degree, he joined the Microsoft. For 17 years, Neukom worked as the Chief Legal officer and general counsel of Microsoft. His duties include the management of the company's governmental and legal affairs as well as its philanthropic activities. He also served as the Executive Vice President of Microsoft.

Since 1995, Bill Neukom has been investing in the San Francisco Giants. From May 2008 until December 2011, he served as the team's Managing General Partner. Under his management, San Francisco Giants had its first win in the World Series since its move to California in 1958.

In 2007 to 2008, Bill Neukom served as the President of the American Bar Association.  With his $850 million net worth, Bill Neukom is currently one of the board members of the Pacific Council on International Policy.
