The grandson of A.N. Pritzker and the son of Jay Pritzker, Daniel graduated from Tufts University in 1981 and earned a Juris Doctor from Northwestern University Law School. As one of the ten billionaire Pritzkers, he and the family sold control of Marmon Holdings, an industrial conglomerate, to Berkshire Hathaway of Warren Buffet in 2007 for a reported $4.5 billion. His grandfather, father, and his uncles Robert and Donald founded Marmon and the Hyatt hotel chain.

As a musician, Daniel Pritzker is the founder, guitarist, and songwriter for the Chicago band Sonia Dada. Aside from being a jazz buff, he is also into filmmaking. He made a silent film Louis about the life of Louis Armstong. It was released in 2010 to a critical praise. His second film Bolden is jazz-induced. It focuses on New Orleans rag-time innovator Buddy Bolden. It was scheduled for release in 2012 but was delayed.

Daniel and his wife Karen are trustees of the Jay Pritzker Foundation. It is a philanthropic fund that founded the Pritzker Challenge at Tufts University with gifts of $10 million and is designed to encourage members of the Tufts community to establish endowed or term scholarships for underprivileged minority students.
