Rodríguez was born as the son of Mexican-American parents in San Antonio, Texas. His mother is a nurse named Rebecca (née Villegas), and his father is a salesman named Cecilio G. Rodríguez. He started his interest in film at age eleven when his father bought one of the first VCRs that came with a camera.

Rodriguez went to the University of Texas at Austin's College of Communication. Here, he developed a love of cartooning. Rodriguez did not have high grades to get accepted into the school's film program. He made a daily comic strip entitled Los Hooligans with the characters based on his siblings. The comic appeared for three years in The Daily Texan, the student newspaper. While doing this, Rodríguez continued to make short films.

Rodriguez does not only write but also direct and produce his films. He also serves as the movies' Steadicam operator, composer, production designer, visual effects supervisor, editor, director of photography, camera operator, and sound editor. That is the reason why people gave him the nickname "the one-man film crew." Rodriguez shot, chopped, and scored the film Once Upon a Time in Mexico and cut and shot the movie Sin City.
