Palmer began her acting career when she appeared in local productions and some commercials while she was studying in elementary. Palmer won a local casting audition, "Search for a Movie Star" in 2003.
After graduating from high school, she received a call from her talent agent, and appeared in the film 2:37. She portrayed the role of Melody, a popular high school student who becomes suicidal after being impregnated by her brother.
In 2:37, she was nominated for the 2006 Australian Film Institute Award for Best Lead Actress for her performance. And the film premiered at the 2006 Cannes Film Festival in "Un Certain Regard" selection.
Palmer starred in the psychological thriller Restraint in 2005.
She made her Hollywood debut with the horror flick The Grudge 2 in 2006.
In 2007, Palmer was set to play the role of villain Talia al Ghul in the DC Comics superhero film Justice League of America. The project was ultimately cancelled by Warner Bros. due to script issues and the 2007–2008 Writers Guild of America strike.

In 2011, she starred in the sci-fi adventure film I Am Number Four. She played the role of Number Six, one of nine aliens hiding out on Earth after their planet was destroyed.
