Born Sarah Anne Callies on June 1, 1977 in La Grange, Illinois, Sarah Wayne grew up in Honolulu, Hawaii. In her youth, she took an interest in acting, and would participate in various school plays at the independent Punahou School. She then attended Dartmouth College after graduating in high school. While studying, she pursued her career in theater. She later went to the National Theater Conservatory in Denver, where she obtained her Master of Fine Arts in 2002.

Callies began her acting career with a guest starring role as Kate O’Malley, a recurring part in Queens Supreme on CBS. Her first lead role was Detective Jane Porter on Tarzan on The WB.
