Born Jordana Ariel Spiro on April 12, 1977 in Manhattan, New York City, New York, she was of Greek and Jewish descent and was raised in Manhattan in the Jewish faith. She has one brother and three sisters. She studied at the Circle in the Square Theatre school and briefly attended the Royal Academy of Dramatic Art in London. She began the MFA Program in Filmmaking at Columbia University in the fall of 2009. She currently splits her time between Los Angeles and New York.

Spiro appeared in the 2009 comedy The Goods: Live Hard, Sell Hard together with Jeremy Piven, Ed Helms and Rob Riggle. This was produced by Will Ferell and Adam McKay, and directed by Neal Brennan. Spiro was credited in The Year of getting to Know Us that premiered at the 2008 Sundance film festival and IFC’s Alone with Her. She also had guest appearances in Cold Case, Out of Practice and CSI:NY. In June 2010, she fell out of the role in the planned 2010-2011 television series Love Bites due to other contractual obligations. She was then casted along with Nicolas Cage and Nicole Kidman in the thriller Trespass. She was a guest star on the Showtime 2011 season of Dexter. She had the lead role in the Fox-TV medical/crime drama The Mob Doctor for the 2012-2013 season. It premiered on September 12, 2012 on the FOX Network.
