The Bollywood king began by starring in series and daily soapies. His first starring role was in Lekh Tandon's 1988 television series Dil Dariya. He played the lead role of Abhimanyu Rai in the series, which depicted a realistic look at army cadet training. This led to additional appearances in Aziz Mirza's television series Circus and Mani Kaul's miniseries Idiot. Khan also appeared in the television serials Umeed and Wagle Ki Duniya, as well as the English-language television film In Which Annie Gives It Those Ones. His appearances in these series drew comparisons to film actor Dilip Kumar's look and acting style, but Khan was not interested in film acting at the time, believing he was not good enough. Khan changed his mind about acting in movies in April 1991, claiming that it was a way for him to cope with the grief of his mother's death.

Khan's Bollywood career was launched with the success of Deewana, for which he received the Filmfare Best Male Debut Award. He has acted in more than 100 movies and earned numerous accolades, including 14 Filmfare Awards. In 2005, the Indian government awarded him the Padma Shri. Both the Ordre des Arts et des Lettres (2007) and the Chevalier Légion d'honneur, the fifth degree of the French Legion of Honour, have been awarded to him by the French government (2014). Khan has five honorary doctorates to his name.

Khan's charitable efforts are focused on health care and disaster relief. In 2011, he received the UNESCO Pyramide con Marni award for his contributions to children's education, and in 2018, he received the World Economic Forum's Crystal Award for his leadership in promoting women's and children's rights in India.

He has established himself as India's most successful actor, owning production companies such as Red Chillies Entertainment and its subsidiaries, as well as Dreamz Unlimited. He paid $75.09 million for the rights to the Kolkata Knight Riders franchise in the Twenty20 cricket tournament Indian Premier League (IPL) and renamed the team Kolkata Knight Riders (KKR). In partnership with Juhi Chawla and her husband Jay Mehta, he owns this team. With a brand value of US$42.1 million in 2009, KKR was one of the richest teams in the IPL.

After six years of dating,  the actor married Gauri Chibber in October 1991. They have three children: Aryan and AbRam, sons, and Suhana, a daughter. Their son, AbRam, was born through the use of a surrogate mother. While Khan firmly believes in Islam, he also values his wife's religion, according to Khan. The actor's family practice both religions, and the Qur'an is displayed alongside Hindu deities in his home.
