At an early age, Khalil Mack played for Pop Warner football. While attending Fort Pierce Westwood High School, he was recruited by high school football coach Waiders Ashmon and joined its football team. During his senior year, he did 140 tackles, eight for a loss, and 9 sacks. He was named third-team All-State and first-team All-Area.

He joined Buffalo Bulls in 2009, and got redshirted during his first year for the team. Mack was named as the MAC Defensive Player of the Year in 2013. He played 13 games with the Bulls, and made a record of 100 tackles, 19 tackles for loss, 10.5 sacks, and three interceptions.

Mack entered the 2014 NFL Draft and selected as the first round, 5th overall pick by the Oakland Raiders.
