After playing college basketball at the University of Florida, Vernon Macklin was selected as the 52nd pick in the second round of the 2011 NBA Draft by the Detroit Pistons. Currently, he plays for the NBA D-League with the Rio Grande Valley Vipers.

In March 2012, he began playing with the Fort Wayne Mad Ants for the NBA-D League. A month after, he returned to the Pistons but in July 2012, he was released which made him a free agent. In August of the same year, he played with the Royal Halı Gaziantep for the Turkish Basketball League. The team released him in November 2012 but a month later, he was absorbed by the Rio Grande Valley Vipers for the NBA-D League.

Last July 2016, Macklin represented the Philippine club Might Sports PH in the 38th William Jones Cup. They eventually won the Gold Medal with an unbeaten record of 8-0.

In March 2018, he signed with the Philippine Basketball Association's Magnolia Hotshots as their import.

In the year 2019, he became part of the Changwon LG Sakers that geared up for the 2019 season of the Korean Basketball League (KBL) with a series of tuneup games against PBA games
