Manetti landed his first television role on the show Chase. He then played Bobby Boyle in Baa Baa Black Sheep. In 1979, he appeared on the NBC series The Duke. 

His role on the television series Magnum, P.I.  brought him fame, airing for eight seasons. He has appeared in feature films, including Emergency!, Tenspeed and Brownshoe, Fantasy Island, Baa Baa Black Sheep, Battlestar Galactica, Renegade, Quantum Leap, JAG, and Walker, Texas Ranger.

In 2011, he and his wife Nancy hosted a weekly radio program on CRN Digital Talk Radio Networks. He joined the cast of the revived version of Hawaii Five-O, playing the recurring role of Nicky 'The Kid' Demarco.
