Nick Fairley started his career as a two-way lineman for the Williamson High School's football team. He was a three-star prospect, and was projected as offensive guard before entering college. He committed to play for Auburn Tigers but could not qualify academically.

In 2007, he played for Copiah-Lincoln Community College. Fairley was a redshirted during his first year with the team. He recorded 83 tackles, and seven sacks in 2008. Then, he transferred to Auburn University where he played for the remaining two years in college. He was a recipient of Lombardi Award, and SEC Defensive Player of the Year in 2010.

Fairley was chosen as the first round, 13th overall pick by the Detroit Lions during the 2011 NFL Draft. He recorded 5.5 sacks, 2 forced fumbles, and 26 tackles in his second seasons with the Lions. On March 13, 2015, he signed a one-year contract with the St. Louis Rams.
